3年前、敏腕刑事と名高い樋口彰吾（唐沢寿明）は愛する妻・未希（菊池桃子）を謎の殺人鬼によって殺されてしまう。
未希は殺人鬼に追われる中必死に110番にかけ、警部補・橘ひかり（真木よう子）が応答するが、一度通話が切れてしまい掛け直したところ、コール音で未希の居場所が殺人鬼に知られてしまい、未希はひかりと電話が繋がったまま殺人鬼に惨殺されてしまうのだった。
1ヶ月後、犯人と思われる人物・相良（平原テツ）が法廷に現れる。検察側の証人としてひかりも法廷に立つが、相良の声は電話で聞いた声と違うため、相良は犯人では無いと証言する。相良が犯人であると確信していた樋口はひかりの証言を聞き激怒し、その場は大混乱に包まれる。
3年後、ひかりは緊急指令室に被害者を迅速に救うための新たなチーム『ECU』を立ち上げる提案を警察幹部にし、更に『ECU』の現場リーダーを樋口に任せたい旨を話す。しかし樋口は愛する妻を失った悲しみや犯人逮捕に至っていない苛立ちからすっかり落ちぶれていた…。
緊急指令室を舞台に、妻を殺された刑事と父を殺された声紋分析官らが通報電話の『声』を頼りに迅速に事件に対応していく姿を描いた『タイムリミットサスペンス』ストーリー。

「ボイス 110緊急指令室」は2017年に放送された韓国ドラマ「ボイス」が原作になっています。
『ボイス』（보이스）は、OCNを運営する韓国のケーブルテレビ「CJ ENM」で放送された韓国のテレビドラマである。日本では『ボイス〜112の奇跡〜』のタイトルでDVD販売、動画配信が行われている。
